ANTIBODY DIRECTED ENZYME PRODRUG THERAPY

In antibody directed enzyme prodrug therapy (ADEPT) an antibody bound enzyme is targeted to tumor cells.This allows for selective activation of a nontoxic prodrug to a cytotoxic agent at the tumor site for cancer therapy.Site-selective prodrug activation results in reduced side effects in remote tissue.This article reviews ADEPT with emphasis on the chemical viewpoint and recent developments.As we know that ADEPT is a type of targeted therapy. It uses a monoclonal antibody to carry an enzyme directly to the cancer cells. First, the monoclonal antibody is given (with the enzyme attached). A few hours later, a pro-drug is given. The pro-drug is an inactive anti-cancer drug. When the pro-drug comes into contact with the enzyme, a reaction takes place which activates the anti-cancer drug. The anti-cancer drug is then able to destroy the cancer cells. As the antibody does not attach to normal cells, the treatment does not affect them.
